Single-use pH Sensors in Biopharmaceutical Applications

Emerson’s Ben Arriola along with Jinbo Hu teamed up to share more about this trend in a BioProcess International special report, Design and Performance of a New Single-Use pH Sensor with Long Shelf Life and High Stability.

In Ben’s article, Introducing the Next-Generation Single-Use pH Sensor, he describes how advancements in single-use sensors have improved quality, reliability, and the level of supplier support.

As part of every good drug manufacturing process, measuring and controlling the critical quality attributes (CQAs) and related critical process parameters (CPPs) are paramount to success — for both drug substances and drug products.

He noted that:

…having a high-performance, reliable pH single-use sensor is critical for continued adoption of SUTs [single-use technologies] in a drug manufacturing facility.

The Rosemount 550pH single-use sensor for in-bag applications has a:

…unique wet-storage technology [which] provides a step change in performance.

In Jinbo’s article, Design and Performance of a New Single-Use pH Sensor with Long Shelf Life and High Stability, he explains that in:

…upstream bioreactor applications, culture medium pH is monitored continuously and controlled within a narrow physiological range. Deviation from that ideal pH range can compromise viable cell concentration, protein productivity, and product quality…

Unlike traditional pH sensors, Rosemount 550pH single-use sensors:

…are compatible with gamma-irradiation sterilization and can be attached as part of single-use bioreactor bag assemblies. By incorporating a unique storage buffer solution, the sensors require no two-point calibration by end users; they can be one-point standardized using the internal storage buffer. The storage-buffer solution maintains the sensor’s glass electrode and reference electrode wet and fresh in storage. That has provided for a long shelf life of two years with outstanding sensor performance including high accuracy, sensitivity, and stability.
